Protonix is a prescription drug that is used to treat and maintain healing of erosive acid reflux disease, also known as erosive GERD (breaks in the lining of the esophagus), and relieve associated symptoms that may include frequent and persistent heartburn and stomach acid backup.

The active ingredient in Protonix (pantoprazole sodium) Delayed-Release Tablets and PROTONIX (pantoprazole sodium) For Delayed-Release Oral Suspension is a substituted benzimidazole, sodium 5-(difluoromethoxy)-2-[[(3,4-dimethoxy-2-pyridinyl)methyl] sulfinyl]-1H-benzimidazole sesquihydrate, a compound that inhibits gastric acid secretion. Its empirical formula is C16H14F2N3NaO4S x 1.5 H2O, with a molecular weight of 432.4.

Protonix (pantoprazole sodium) is supplied as a delayed-release tablet for oral administration, available in 2 strengths (40 mg and 20 mg); and as delayed-release granules for oral administration, available in the 40 mg strength.

Each Protonix Delayed-Release tablet contains 45.1 mg or 22.6 mg of pantoprazole sodium sesquihydrate (equivalent to 40 mg or 20 mg pantoprazole, respectively) with the following inactive ingredients: calcium stearate, crospovidone, hypromellose, iron oxide, mannitol, methacrylic acid copolymer, polysorbate 80, povidone, propylene glycol, sodium carbonate, sodium lauryl sulfate, titanium dioxide, and triethyl citrate.

The recommended adult oral dose is 40 mg given once daily for up to 8 weeks. For those patients who have not healed after 8 weeks of treatment, an additional 8-week course of Protonix may be considered.

Protonix Delayed-Release Tablets are prepared as enteric-coated tablets so that absorption of pantoprazole begins only after the tablet leaves the stomach. The enteric-coated pill has a special coating to protect your stomach. Breaking the pill could damage this coating. Do not crush, chew, or break an enteric-coated tablet. Swallow the pill whole, with or without food in the stomach.

Protonix Delayed-Release Oral Suspension is bioequivalent when administered orally in applesauce or apple juice, or mixed in apple juice and administered through a nasogastric tube. Protonix Delayed-Release Oral Suspension should be taken approximately 30 minutes before a meal.


Do not stop taking Protonix even if you begin to feel better. Your symptoms may improve before the condition is fully treated.


In clinical trials, the most frequently reported side effects with Protonix Delayed-Release Tablets were headache, diarrhea, and gas. Relief of your symptoms while on Protonix does not exclude the possibility that serious stomach conditions are present. Patients who are allergic to any ingredient of Protonix should not take it. Get emergency medical help if you have any of these signs of an allergic reaction:
  • hives;
  • difficulty breathing;
  • swelling of your face, lips, tongue, or throat.


  • Pantoprazole is not removed by hemodialysis. In case of overdosage, treatment should be symptomatic and supportive.

    Some conditions must be treated long-term with Protonix. The chronic use of Protonix has caused stomach cancer in animal studies, but it is not known if this medication would have the same effects in humans. Talk with your doctor about your specific risk of developing stomach cancer.

    This medication is not expected to be harmful to an unborn baby. Tell your doctor if you are pregnant or plan to become pregnant during treatment. It is not known whether Protonix passes into breast milk or if it could harm a nursing baby. Do not use this medication without telling your doctor if you are breast-feeding a baby.

    There are no restrictions on food, beverages, or activity while taking Protonix, unless otherwise directed by your doctor.

    Keep this medication in the container it came in, tightly closed, and out of reach of children. Store it at room temperature and away from excess heat and moisture (not in the bathroom). Throw away any medication that is outdated or no longer needed. Talk to your pharmacist about the proper disposal of your medication.
